The character Pannacotta Fugo is able to evolve his stand Purple Haze into Purple Haze Distortion by entering resolve mode or by using a Great Heat Attack. This is notable as Purple Haze Distortion actually made its debut in the light novel spin-off Purple Haze Feedback which was not written by series creator Hirohiko Araki and is thus considered non-canonical.

CyberConnect2 president and CEO Hiroshi Matsuyama explained that JoJo's Bizarre Adventure: All-Star Battle was conceived "partly thanks to" the 2006 video game JoJo's Bizarre Adventure: Phantom Blood. When that game was finished, the developers came up with the idea of a new game involving all the story arcs of the series.

Giorno Giovanna's HHA move has unique animations for certain characters in the game, five of which reference various panels from the manga Jojo's Bizarre Adventure and/or abilities of the person being attacked:
• When used against Jonathan Joestar, Giorno's stand will apply its thumb onto Jonathan's eye, referencing Part 1 (Phantom Blood) in which Dio Brando challenges Jonathan Joestar to a boxing match and cheats by jamming his thumb into his eye, causing it to bleed and temporarily blind him.
• When used against Jean Pierre Eiffel (Polnareff in Japan), Jean's tongue will stick out, referencing to Part 3 (Stardust Crusaders) in which another character, Hol Horse, is pointing a gun towards him and Jean is trying to warn the rest of his group about it by pointing behind him with his tongue.
• When used against DIO (the mononymous Part 3 version of Dio Brando), the attack hits his upper-head. Normally the attack hits other characters in the cheek or jaw, but this is likely a reference to Jotaro Kujo managing to successfully attack DIO, temporarily caving in his skull.
• When used against Bruno Bucciarati, his face will contort more so than any of the other characters, in reference to Giorno and Bruno's first fight in Part 5 (Vento Aureo), which was also the first time Golden Wind (Gold Experience in Japan) was used offensively.
• When used against Father Enrico Pucci of Part Six (Stone Ocean), Golden Wind will hold Pucci's cheek in one hand and punch the other cheek with the other, referencing Spoiler:his death at the hands of Emporio Alnino, who pins his face to the floor while inside Burning Down (Burning Down the House in Japan) before crushing his skull with a volley of punches.

The English translation of the game involves a large number of name changes to many characters as well as stand names for those that have stands, all to avoid any potential lawsuits with musicians/bands. Some examples are the character "Vanilla Ice" being changed to "Cool Ice", Giorno's stand "Gold Experience" was renamed to "Golden Wind" and the character "Jean Pierre Polnareff" was changed to "Jean Pierre Eiffel."
The original Japanese voice acting will still say the original names, despite the subtitle changes.

When Ikuro Hashizawa has a large amount of health, his eyes are blank white and his mouth does not move. As a result he doesn't emote when struck by Giorno's HHA.
When he is low on health, pupils appear in his eyes and his mouth will move in attack and taunt animations, and he visibly shows pain in Giorno's HHA.
This mirrors Ikuro's growing control over the Baoh parasite in the original Baoh manga.

The game also has unique Great Heat Attack animations depending on who's in battle.
If Jotaro is hit with Part 3 DIO's GHA move, Jotaro's Star Platinum will attempt to punch the streamroller away. If the Jotaro player inputs Jotaro's time stop here, a special animation plays of Jotaro stopping time and escaping, whilst also minimizing the damage he receives.
If Funny Valentine is hit by Johnny Joestar's GHA, an extra animation plays where Valentine attempts to use his Love Train barrier only for Johnny's Tusk ACT 4 to force it open and thrash Valentine. Throughout this, Johnny whispers heartfelt thanks toward his friend Gyro.
If Valentine is hit by Gyro Zeppeli's GHA, Valentine again throws up Love Train only for Gyro's Ball Breaker Stand to rip through it. Gyro rides past Valentine on his horse, and Valentine is trashed by the Super Spin energies left in Gyro's wake.

Certain characters in the game have special intro dialogue with their past self should they be pitted against one another. This includes Dio's Part 1 and 3 selves, Yoshikage Kira, Kosaku Kawajiri and both versions of Joseph Joestar.
Notably, Kosaku will wonder if his encountering his past Kira identity is an effect of his Bites the Dust ability.

In the English version of the game, if Joseph Joestar beats the DLC character Lisa Lisa, his victory screen has him saying "Why did it have to come to this?! You are tearing me APART, Lisa Lisa!"
This quote is a joke-translation using the same quote from the movie The Room in which Tommy Wiseau's character yells out "You are tearing me APART, Lisa!"

Joseph Joestar has a unique counter-taunt, where he predicts his opponent's next line, which they proceed to say completely, and then react in surprise. He has a different version for each of the 41 characters, which includes two versions of himself.
Using this counter-taunt against Dio Brando will make Joseph say "Next you will say 'WRYYYYYYYYYY'", with Dio responding as expected. The "WRYYYYYYYYYY" that Joseph mimics sounds similar to the synthesized version of the yell Dio Brando makes in the Capcom-made arcade fighter JoJo's Bizarre Adventure, which became a semi-popular internet meme that was parodied in several videos.

In 2023, the character Leone Abbacchio was added to JoJo’s Bizarre Adventure: All Star Battle R as a DLC character over ten years after the original game’s release. Despite being a main cast member of the fifth part of JoJo’s Bizarre Adventure, he was previously only playable in the 2002 Japan-only PlayStation 2 game Jojo no Kimyou na Bouken: Ougon no Kaze, due to the developers having trouble trying to make his ability Moody Blues, which causes his opponents to replay their moves, viable for combat. This led to him becoming a very highly-requested fighter for the game before he was eventually added.
