During Act 3 in the original release of the game, Spoiler:Monika would make a short reference to the Super Smash Bros. series of games. She says the following sentence. "Hey, have you heard of a game called Super Sma--". She then interrupts herself and continues the conversation. This dialogue was removed in the Version 1.1.1 update, likely because a following line encouraged players to contact developer Dan Salvato (who is also Spoiler:a prominent member of the Super Smash Bros. community), which became a risk once Doki Doki Literature Club! achieved widespread popularity.

In Metropolis, if you get through the whole level without beating a single enemy and talk to the Inventor Droid at the end, she'll say "I've Successfully developed a super powerup here Spyro... knock off just a few more enemies and you can be the first ever to use it." This dialogue isn't normally available to see or hear because the power up only requires one enemy to be defeated to work and there are enemy pigs that self destruct when you get close enough to them.

Attachment Two P's name and design are a tribute to the second player character from the Capcom arcade shooter Forgotten Worlds. Additionally, his backstory references this as well, as the players start out shirtless, and his catchphrase is an exact quote from the ending of the game.

On a side note, Two P is skilled at handling heavy weapons, referencing the Unknown Soldiers' large guns in Forgotten Worlds.

When the player's creature evolves into the tribe stage, a cutscene will play in which the creature learns how to use a stick as a weapon, then, as a way of celebration, throws the stick into the air and it lands on the creature's head.

This cinematic sequence is a parody of the opening sequence from the Stanley Kubrick film 2001: A Space Odyssey, where an ape learns how to use a bone as a weapon. Additionally, the intro cutscene from Spore features a remix of 'Sunrise', which is the initial fanfare of the tone poem "Also Sprach Zarathustra".

• On New Years Day (01/01/XXXX), while playing as Joey T., he will wear a Happy New Year's baby costume, and the boxing ring has a design that reads "Happy New Year".
• On Valentine's Day (02/14/XXXX), Lulu Valentine sports a sexy lingerie costume along with the ring being designed with roses and the words "Happy Valentine's Day".
• On St. Patrick's Day (03/17/XXXX), the boxing ring has a green texture designed with four-leaf clovers, along with the words "Happy St. Patrick's Day" and having the referee dressed as a leprechaun.
